Inhibitory Effects of Isoderrone on Platelet Aggregation through Regulation of Cyclic Nucleotides

Abstract
Extract of Cudrania tricuspidata is commonly used in ethnomedicine throughout Eastern Asia, and various studies have reported that Cudrania tricuspidata has physiological activities such as anti-diabetes, anti-bacterial, and anti-cancer effects. In addition, it has been reported that root extract of Cudrania tricuspidata has anti-platelet effects. Therefore, we studied the potential efficacies of this natural compound by focusing on the inhibitory effect of isoderrone from Cudrania tricuspidata on human platelet aggregation. Collagen-induced human platelet aggregation was dose-dependently inhibited by isoderrone and it suppressed collagen-induced elevation of [Ca2+]i mobilization. In addition, isoderrone inhibited collagen-induced fibronectin adhesion to ¥á¥±b/¥â3. Thus, in the present study, isoderrone showed an inhibitory effect on human platelet activation, suggesting its potential use as a natural substance for preventing platelet-induced thrombosis.
